Chimera Germline Testing: The chimeras are setup for germline testing when they are 8 weeks old. 4-5 chimeras will be setup with CD-1 females, Rosa26-FLP females for CKO or hprtCre females for mutation constructs at the same time. Once germline transmission is confirmed, we would also have F1 mice with neo cassette removed. We'll develop PCR primers and PCR conditions for F1 genotyping. For subtle mutation knockin construct at least two F1 mice will be sequence. 8-10 F1 mice will be sent to invevstigator. The fee for germline testing is $800 that includes the mice (CD-1, FLP, Cre or C57BL/6) used for matings, PCR genotyping method and genotyping of F1 mice. The fee doesn't include per diem costs charged by the Animal Care Facility.
